Masterson of Kansas is set in the volatile 1870's town of Dodge City in Kansas. Sheriff William Barclay 'Bat' Masterson (George Montgomery) and Wyatt Earp (Bruce Cowling) are longtime friends. This friendship results in a truce between the Sheriff and convicted gambler John Henry 'Doc' Holliday (James Griffith). Bat also agrees to travel to Indian territories in order to extradite a certain Amos Merrick (John Maxwell), who is a suspect in the murder of an Army colonel. The Sheriff seeks out an Indian chief called Yellow Hawk (Jay Silverheels), whose tribe is sheltering Amos, but Hawk seems reluctant to hand over Amos. The Indian tribe supposedly has proof that Amos Merrick is innocent and they are bent to prove it, even though countless testimonies are pouring in. Wyatt Earp and 'Doc' Holliday also get involved in the manhunt even though Holliday has to be persuaded by Amos' daughter Amy Merrick before he agrees.
