Sheriff of Wichita is the story of Lt. Raymond D'Arcy (Clayton Moore), a United States cavalry officer accused of stealing a $25,000 payroll. Insisting he's innocent, D'Arcy engineers an escape from prison to search for Major Bishop, his missing superior officer, the only man who can exonerate him of the theft. Bishop's daughter Nancy (Lyn Wilde) refuses to believe her missing father had any involvement in the theft. Determined to prove his innocence, Nancy gathers five of her father's colleagues at an abandoned army fort - the same fort where fugitive D'Arcy is hiding out. The Sheriff of Wichita, Sheriff Rocky Lane (Alan Lane), gets news of the gathering and infiltrates the group, posing as one of the invited men. Meanwhile, a group of local bandits led by the murderous Sam Stark (Roy Barcroft) have also heard of the story and stealthily descend upon the fort. The truth is exposed when these two groups explosively collide.
