Kaji makes a trip to Kyoto for a secret meeting, as Asuka goes out on a blind date and Shinji and Rei stay after class to clean up. The next day Shinji visits the grave of his mother with Gendō, the first time in 3 years. Misato, Ritsuko and Kaji attend the wedding of a friend and reminisce about their past, while Asuka and Shinji find something to pass the time.

In Central Dogma, where Rei is encased inside a glass tube full of Link Connection Liquid (LCL), the letters A,C,T, and G form two rows that combine and circle the huge area. The letters are abbreviations for the four nitrogen bases that make up Deoxyribonucleic Acid or DNA: Adenine, Cytosine, Thymine, and Guanine.

At the end of this episode, it is revealed that Rei in Unit-00, at the end of Episode 14, was carrying the giant forked spear to place it in the chest of the great white behemoth that resides in Terminal Dogma.
