The mayor's cameo on the ABC daytime drama is scheduled to air in the US Wednesday.
ABC says the mayor, playing himself, will meet Susan's character Erica Kane at a hot spot.
Villaraigosa filmed the scene last month to welcome the show, which moved production from New York to Los Angeles.
He's not the first LA mayor to go Hollywood.
Sam Yorty, Tom Bradley and Richard Riordan all made guest appearances on TV shows.
