)Well, the Hello Nurse Song does say that Wakko is 7, but the creators have said that isn't canon.
I always figured Dot was around 6. That's why she's so cute and precocious.
Wakko, about 8-9. He just seems to be at that age, filled with boyish enthusiasm.
Yakko, on the other hand, could be anything from 10-13. But I always figured he was 13, since he was so obsessed with girls. You know, teenage hormones.
That's my take on it, anyway.

No, Dot is definitely younger than Wakko. He has called her his little sister in the show before.
And in "Take My Siblings, Please" Dot even tells the troll to eat her brothers because she is so small and they are way meatier than she is.
Also in Wakko's Wish, Wakko says "I'd wish for an operation for my little sister Dot." Dot's definitely the cute younger sister. Though exactly how much younger than Wakko is undetermined.
Yeah, she does act a bit older sometimes, but everyone knows girls are more mature at that age.
According to the "Animaniacs Bible", the original plan was for them to be teenagers. But since the Warners seem to display traits of younger children more so than teens, they could have changed their mind about that as the show progressed, which may be why a definite age was never given, probably deciding that it would be better that the audience draw there own conclusion toward their ages. The attraction toward a pretty girl or in Dot's case a handsome boy seem to be the only thing real teen trait that was seen in them. The most likely teen canidate in the Warners would be Yakko, not just because he's the oldest, but because he seemed to show more knowledge of things beyond what children should know ( "Goodnight Everybody!" ). But still the child nature shows a dominance in him as well. Although the most accurate, some things in the animaniacs bible never completely went through, apparently the nurse was supposed to just be some hot nutcase who thought she was a nurse.....
