When Bobby is attacking Dean and Sam with his knife, his grip shifts from a regular grip to a reverse grip several times as the camera shot changes.
Eve: God doesn't care about you.
Rick: Sure He does.
Eve: Your Father made you and then abandoned you. So you pray. You see signs where there's nothing. But truth is, your Apocalypse came and went, and you didn't even notice. A mother would never abandon her children like He did. You'll see.
Bobby: So what?
Rufus: So we partnering on this or not? Come on, it's not rocket surgery. We're here, lets do this. Just like old times.
Bobby: As long as I get to drive.
Rufus: Hell, no.
Sam: It really is good to see you, Rufus.
Rufus: I can believe it. It must get old dealing with this miserable cuss here all by yourselves.
Sam: Is it that obvious?
Bobby: Why don't you three get a room?
Bobby: So... you're Samuel.
Samuel: You must be the guy pretending to be their father.
Bobby: Somebody ought to.
Dean: I'm not in the mood. I just had a 12" herpes crawl out of my ear.
Dean: You heard me.
Sam: You saying he wasn't a monster when I ganked him?
Rufus: One way to find out. Bobby, you got a cranial saw in the car?
Bobby: Of course.
Rufus: And I want you and you... okay, I want you and you to watch him and him... all right, if anything crawls out of anybody, step on it.
Sam: What he did to us... but...
Dean: There's a but?
Sam: I can't help but think , what would Mom say?
Dean: You know what I think Mom would say? She'd say, "Just because you're blood doesn't make you family. You got to earn that."
Rufus: (to Dean, Sam, Bobby) This can't be the afterlife because the three of you are here.
Dean: Hey there, you little herpes.
Sam: Why do you keep talking about herpes?
Dean: What? I don't. Shut up. Shut up!
Dean: Who is she, this Eve bitch?
Slug: The Mother of All of us. And the end of all of you. By the time she's done, there'll be more creatures than humans. You'll live in pens. We'll serve up your young and call it veal.
Bobby: It was Omaha. It was my fault. And he never let it go.
Dean: Well, he should have.
Bobby: You don't know what I did, Dean.
Dean: It doesn't matter.
Bobby: What do you mean, it doesn't...?
Dean: I mean, at the end of the day, you two are family. Life is short, and ours are shorter than most. We gonna spend it wringing our hands? Something's going to get us, eventually. And when my guts get ripped out, just so you two know, we're good. Blanket apology for all the crap that anybody's done, all the way around.
Sam: Some of us pulled a lot of crap, Dean.
Dean: Well, clean slate.

Injoke: One of the names on Sam's cell phone is Chris Cooper. Chris Cooper is the show's prop master.
Referencing the 1939 novel by mystery author Agatha Christie. In the book, ten people find themselves stranded on an island and realize that one of them is a murderer trying to eliminate the other nine. Each person dies in a manner similar to the children's nursery rhyme, "Ten Little Indians." At the end, all ten individuals are apparently murdered, leaving the case unsolved except for a postscript when the murderer's explanation is provided.
Dean: It's like a Khan worm on steroids.
Referencing Star Trek II: The Wrath of Khan. The title villain places larval Ceti eels in the ears of those he wishes to interrogate. The larva eventually kill the host, although in a few cases they will leave the victim on their own.
Samuel: Just because you're Dr. Jekyll at the moment doesn't mean you get all high and mighty.
Referencing, The Strange Case of Dr. Jekyll and Mr. Hyde, a story by Robert Louis Stevenson, originally published in 1886. In the tale, Dr. Jekyll, a peaceful man, created a potion that unleashed his "evil side", the murderous Mr. Hyde. Since its publication, the term "Dr. Jekyll and Mr. Hyde" has come into colloquial use, to refer to the good and evil parts of a person.
Rufus: All right, let's play Operation.
Referencing the Milton-Bradley board game (now manufactured by Hasbro) where players attempt to remove small plastic pieces from a picture of a patient, Cavity Sam, using a pair of metal tweezers. The edges of the holes are lined with metal, and grazing them with the metal tweezers sets off Cavity Sam's red light-bulb and a buzzer noise. The game has spawned numerous variants, including patients such as Buzz Lightyear, Iron Man, and a Dalek.
Dean: No way out now, Sluggo.
Referencing The Mr. Bill Show, a series of skits on Saturday Night Live, and Mr. Sluggo, the villainous clay figure that torment the hapless Mr. Bill. Sluggo rarely gets to do much, since Mr. Hands is the one who ends up inflicting various cruel fates on Mr. Bill.
