In a Laramie hotel room Jim lies dying from a gunshot wound and will not let his sweetheart, Clara Malone (Shirley Knight), go for medical aid. He tells her the money is hidden in an old fort near Medicine Bow and dies. She packs her things and heads off down the street but runs into Trev Holcomb (Simon Scott) and Sam Barton (John Kellogg) who want to know were Jim is to be found. She tells them he is dead and they figure she knows where the money is and say she better tell them or she will get what Jim got. Clara manages to get away, fakes a suicide and later boards a night stage towards Medicine Bow, the only other passenger on which is the Virginian. The stage loses a wheel and plunges down a hill killing the driver. The lamp from the stage sets the grass on fire and threatens to burn Clara but she is rescued unconscious by the Virginian who takes her to nearby Shiloh Ranch. She wakes up with amnesia and is nursed by Betsy. Meanwhile, Trev and Sam do not believe she is dead and start searching surrounding towns.

The Virginian nicknames Clara as April McComber and when she has recovered sufficiently he gets her a room with Martha Winslow (Monica Lewis), the Medicine Bow school teacher. There is talk of hiring "April" as a second teacher and romance blossoms between her and the Virginian. Meanwhile Sam arrives in town and sees Clara at a local dance. He dances with her but is confused when she shows no signs of recognition. He wires Trev who arrives and visits Sheriff Brannan (Harlan Wade). He pretends he is a Cal Harris who is looking for his half sister who had written him in Salt Lake that she was coming but is much overdue. He is told there is a woman amnesiac in town and the Sheriff, upon seeing Clara's photo, confirms that is her. Clara does not recognize Trev but posing as her brother he arranges to take her to Salt Lake to see a specialist about her memory problems.

The next night as the Virginian is leaving Clara, she accidentally sets a curtain on fire at Martha's house and in fright, passes out. They send for her brother but before he arrives Clara wakes up with her memory regained. She can not remember what has happened since the stage accident but overhears the Virginian and Martha talking about her outside her bedroom window and learns of the memory loss. Trev arrives and she recognizes he has been posing as her brother. She lets on to no one however, that she has regained her memory. The next morning after Martha has left for the school she begins packing to escape. However, Trev and Sam arrive and although she pretends to still be "April" they discover she has regained her memory. She tries bargaining with them, saying she wants to stay in Medicine Bow and that in return for their silence she will tell them where the money is hidden.

The Virginian then arrives but is surprised by Trev and Sam. They reveal who Clara really is and that Jim was a fellow bank robber who had double-crossed them. The two men take the Virginian and Clara with them to make sure she is telling the truth about where the money is hidden. They have to drive through town in a buggy and encounter the Sheriff. The Virginian says he is taking "April" and her brother back to Laramie and passes on a cryptic message for Trampas to make arrangements for his absence from Shiloh. After they ride off Trampas arrives and gets the message from the Sheriff. He manages to deduce that something is wrong and that the Virginian and the others are heading for the abandoned military fort. So Trampas and the Sheriff ride off in pursuit. Meanwhile, the others arrive at the fort and Sam discover the money. The Virginian manages to knock Trev's gun out of his hand but would have been shot by Sam if not for the timely arrival of Trampas who kills Sam with a rifle shot. The Virginian then beats up Trev, knocking him unconsious. The Virginian sees off Clara at the train as she is going to see her family whom she has not seen in five years. However, she hints she will return.
